What types of things do you think should be included in your mobile home insurance coverage? The reason why the answer to this question is not as simple as it might appear is because while a mobile home in many cases is just like a traditional home, there is no land ownership covered in a mobile home insurance coverage, which means that particular care needs to be taken in respect to physical damages and liability coverage.

The coverage you have might only extend to the tree. Because you do not own the land that the visitor was standing on when he got hit by the tree, in theory your liability claim may not extend to that action. However, by law, because the damage sustained by the visitor was caused indirectly by your mobile home, you could still be sued.

    Probably the most common transcription factor binding domain is the helix turn helix (HTH). Funny enough I spent a year working on a Leucine Zipper transcription factor, but I am most familiar with HTH, so I will use that as an example.

    As far as dimerization is concerned, the binding site or binding element is always located on two major grooves, yet a HTH domain can only fit into one groove. So what ends up happening is that the HTH proteins dimerize and fill each grove. So if it didn't dimerize, the binding would have a very low affinity.

    Now, in reference to specificity, since you need both binding (regulatory) sequences present in the DNA strand, the specificity is increased with the need to dimerize. For example, it may be common to find one sequence, but to find the sequence in two major grooves next to each other…well that's much more rare.

    Interestingly enough, the two proteins are always inverted in reference to each other. If you hold your thumb and index finger apart, it will resemble one. Now make this with both hands and put them parallel to each other pointing at each other with both thumbs facing down. The index fingers would bind the major grooves while the thumb wraps around the sugar phosphate bond.

    This is a good picture showing you how both the domains bind a major groove, but its tough to see how they're inverted. So when looking at this picture, imagine the one on top as the N end facing you while the bottom one has the C end facing you.

    Now, the second part of your question asks what they can interact with.

    Well as I already mentioned, only major grooves interact. Also each specific transcription factor has a unique amino acid sequence on its "index finger". This region is called the recognition sequence. So really an HTH domain can interact with any sequence as long as it matches the recognition sequence on the protein.
